چکیده
T'b is comilitmication emphasizes the nieed for conltin1uouts conitrol of pH when macerating acidic plant tissues for the l)url)ose of isolating tightly coupled mitochondria. A useful device for such control is described. Mitochonidria capable of oxidative but not pl)osphorylative activity have been isolated fromli apple fruits wlhenl either alkaline homogeniates (5) or l)oIyvinylpyrrolidone (PV7P) (1) was used to supl)ress phenolic compounds. However, it was niot until a pH slightly below 7, pluis 0.5 % I'VP, wvere used that phospliorylating, "coupled" miitoclhonldria were obtained froml the l)arenchymatous tissues of both pear andl apple fruits (7, 8). Loomlis and Battaile (6) have sinice slioNvni that PVP is less effective at an ailkalinie pH. Wiskich (9) utilized an isolation medium at pH 7.4, btut containing 1 % PVP, to obtain apple mitochondria with respiratory colntrol ratios (RCR) of somewhat less than 2. Hulmle ct al. (2) reported results similar to those obtainied by Wiskiclh. Tightlty coupled mitoclhondria (RCR's approachilng infiniity) were obtained from pear tissues only wlhen additional precautions were taken to control pH during nmaceration. This was accomplished with the device illustrated in figures 1 and 2. The surface of the stainless steel screen (fig 2) has been grotund slightly to provide shearing edges. Downward pres-

Necessary conditions for isolation of tightly coupled higher plant mitochondria.
A number of parameters associated with the isolation of plant mitochondria were carefully examined in the hope of optimizing the isolation technique. Dark-grown mung bean (Phaseolus aureus var. Jumbo) hypocotyls were used as the source of mitochondria. Necessary conditions for the isolation of tightly coupled plant mitochondria include: (a) gentle tissue disruption; (b) rigorous exclusion of co...
متن کاملProperties of Higher Plant Mitochondria. I. Isolation and Some Characteristics of Tightly-coupled Mitochondria from Dark-grown Mung Bean Hypocotyls.
The mitochondria isolated from dark-grown mung bean hypocotyls oxidize succinate, l-malate, and externally added reduced nicotine adenine dinucleotide (NADH) with good respiratory control. While the pattern of respiration resembles that of animal mitochondria, there are 4 basic differences between the respiratory properties of mung bean and animal mitochondria: A) the ability to oxidize NADH, B...
متن کاملIsolation and oxidative properties of intact mitochondria isolated from spinach leaves.
A procedure was described for preparing intact mitochondria from spinach (Spinacia oleracea L.) leaves. These mitochondria oxidized succinate, malate, pyruvate, alpha-ketoglutarate, and NADH with good respiratory control and ADP/O ratios comparable to those observed with mitochondria from other plant tissues. Glycine was oxidized by the preparations. This oxidation linked to the mitochondrial e...
متن کاملIsolation of intact plastids from a range of plant tissues.
A technique for the isolation of intact plastids from spinach (Spinacia oleracea) and pea (Pisum sativum) leaves, pea roots and castor bean (Ricinus communis) endosperm is described. This technique involves brief centrifugation of whole homogenates on density gradients. Intact plastids were located in the gradient by assaying for triose phosphate isomerase activity. Contamination of the plastic...
متن کاملMetabolism of sea urchin sperm. Interrelationships between intracellular pH, ATPase activity, and mitochondrial respiration.
Mitochondria of sperm of the sea urchin Strongylo-centrotus purpuratus are tightly coupled before induction of the acrosomal reaction. When the sperm are diluted at low external pH (pH 5.5) or in high potassium (200 mM) or in the absence of sodium, their internal pH is acidic (6.2-7.0) as measured by amine accumulation. Under these conditions the internal ATPase activity (primarily the dynein A...
